ABSTRACT

Objective@#To explore the relationships between the characteristics of childhood sexual abuse and non-suicidal self-injury in nursing female college students.@*Methods@#Two medical colleges and junior colleges were selected in Anhui province. A total of 2 549 female nursing students in grade 1 to 3 were asked to fill a questionnaire regarding sociodemographic information,childhood sexual abuse and non-suicidal self-injury.@*Results@#The reported rate of non-suicidal self-injury among female nursing students in the past six months was 8.2%. Sexual abuse at any time during childhood (primary school or earlier,middle school and high school) increased the risk of non-suicidal self-injury among female nursing students (P<0.05). Exposure to sexual abuse in all three periods was associated with 5.04(95%CI=1.73-14.62) times odds ratio than that of those who not exposed to sexual abuse (P<0.01). Only contact sexual abuse and both contact and non-contact sexual abuse in childhood were correlated with nonsuicidal selfinjury among female nursing students [OR(95%CI)=2.21(1.48-3.29), 3.56(2.13-5.96)] (P<0.05). Two patterns of sexual abuse experiences were identified,including persistent sexual abuse (3.1%) and the other group is occasional sexual (96.9%). Persistent sexual abuse in childhood was correlated to higher risk of non-suicidal self-injury compared with occasional sexual abuse (OR=2.61,95%CI=1.35-5.05,P<0.01).@*Conclusion@#The occurrence periods,types and patterns of sexual abuse in childhood are closely related to non-suicidal self-injury in female nursing students.

ABSTRACT

Objective:To explore the prognostic evaluation value and influencing factors of prognostic nutritional index (PNI) in elderly patients with stroke, and to analyze the value of PNI as an indicator of stroke outcome and prognosis.Methods:The clinical data of 356 elderly patients with cerebral apoplexy treated in hospital from December 2015 to June 2019 were analyzed retrospectively. According to PNI index, there were 154 cases of PNI≥45 and 154 cases of PNI.Results:General data showed that patients with PNI < 45 had higher age, body mass index (BMI), TP, ALB, lymph, TG, Glu, BMI and Mrs scores than those with PNI≥45, The difference was statistically significant ( χ2 values were 0.005-0.083, t values were 0.037-1.455, P<0.05). The results of multivariate analysis showed that PNI ( HR value was 1.43, 95% CI 0.94-2.25 , P<0.01, standardized partial regression coefficient was 0.051) had the highest contribution and was an independent risk factor for prediction of postoperative risk. Moreover, kappa coefficient indicated that PNI was consistent with MRS ( P<0.01). Conclusions:PNI is an independent factor affecting the prognosis of elderly patients with CS, which is of great significance to the evaluation of prognosis and analysis of influencing factors in elderly patients with CS.

ABSTRACT

Objective@#To investigate the correlation between childhood maltreatment and quality of life (QOL) in nursing girls in Anhui Province, so as to provide reference for improving QOL of nursing girls.@*Methods@#The stratified cluster sampling method was used to select students of grade 1 to 3 in nursing major from 2 colleges and 2 specialized colleges by self-administered questionnaire in Anhui province. The questionnaire was conducted to analyze the relationship between childhood abuse and neglect and QOL of female nursing students. Multiple linear regression model was used to assess the associations between childhood maltreatment and QOL.@*Results@#Among 2 549 female nursing students, the mean scores of 4 dimensions of QOL were physical (12.85±2.01), psychological (13.86±2.40), social relationship (12.83±2.84) and environment (13.37±2.21). The scores of QOL among college students were higher than specialized college students, high grade students were higher, and those who have a good relationship with their parents were higher(P<0.01). The prevalence rates of childhood physical abuse, emotional abuse, sexual abuse, physical neglect, and emotional neglect were 4.9%, 10.6%, 7.3%, 6.7% and 4.1%, respectively. The scores of QOL were lower among female nursing students with childhood maltreatment than those without childhood maltreatment(P<0.01). The results of multiple linear regression analysis showed that the number of childhood abuse and neglect were negatively correlated with physical, psychological and environmental dimension of QOL, and the number of childhood maltreatment were significantly dose-responsive to quality of life(P<0.05).@*Conclusion@#The female nursing students are still greatly neglected, and the experience of childhood maltreatment is negatively related to the quality of life among female nursing students.

ABSTRACT

Objective To investigate whether myricetin (Myr) exert protective effects on acute lung injury induced by LPS and to explore its underlying mechanisms. Methods A total of 72 mice were divided into the control group, model group, myricetin group and active group, 18 in each group. Male BALB/c mice of the myricetin were given an intragastric injection of Myr (50 mg/kg), and the active group dexamethasone (Dex, 5 mg/kg) 1 h before intranasal instillation of LPS. While the other groups were given the same volum saline. 24 hours after LPS exposure, mice were sacrificed and various samples were collected for further analysis. Both levels of tumor necrosis factor-α (TNF-α) and interleukin-1β (IL-1β) in the bronchoalveolar lavage fluid (BALF) were measured by ELISA kits. The numbers of total cells, macrophages, and neutrophils in the BALF were also determined using differential leukocyte cell count method. The phosphorylations of nuclear factor-kappa B (NF-κB) and IκB were further detected by western blot analysis. Results Compared with the model group, the levels of macrophages and neutrophils in BALF in the myricetin and active groups significantly decreased (P<0.01), while the total protein and W/D ratio of the lung tissue significantly decreased (P<0.05). Compared with the model group, the levels of TNF-α (5.47 ± 2.54 pg/ml, 4.19 ± 0.50 pg/ml vs. 15.28 ± 1.27 pg/ml), IL-1β(336.20 ± 41.06 pg/ml, 247.99 ± 25.12 pg/ml vs. 464.88 ± 34.51pg/ml), the MPO (0.74 ± 0.12 U/g, 0.61 ± 0.08 U/g vs. 1.66 ± 0.07U/g) in the myricetin and active groups significantly decreased (P<0.01). Compared with the model group, the expression of p-IκB/IκB (0.61 ± 0.01, 0.59 ± 0.01 vs. 0.95 ± 0.01), p-NF-κB/NF-κB (0.54 ± 0.01, 0.59 ± 0.03 vs. 0.96 ± 0.02) of the lung tissue in the myricetin and active groups significantly decreased (P<0.01). Conclusions Collectively, these findings revealed that Myr may alleviate the LPS-induced acute lung injury via relief of inflammatory reactions associated with blockage of the NF-κB signaling pathway.

ABSTRACT

Objective To obtain highly purified astrocytes and identify the cells in each stage to support further studies.Methods The cerebral cortex of a neonatal SD rat was isolated and prepared into single cell suspension.The obtained cells were purified by differential adherence and shook at a constant temperature.By inverted phase contrast microscopy and HE staining,cell morphology was observed.The immunofluorescence staining with anti-mouse GFAP was used to identify the cells.Results The primary cortical cells developed rapidly at 3 d after culture and covered the flasks at 9-12 d.At this time,the cells showed stratification and the astrocytes lay at the lower layer.GFAP positive rate was only about (67.2 ±7.1)%.After the first passage,GFAP positive rate increased obviously (84.0±6.0)%. However, oligodendrocytes and microglias could not be removed completely,and the cells also showed stratification.Through 3 times of passages,we obtained many single species of astrocytes showing satellite shape with 2 or 3 processes,big cell body and round or oval-shaped nuclei leaned to one side.Immunofluorescence staining showed that nearly all of the cells were strong positive and the positive rate reached as high as (97.6 ± 2.4 )%.Conclusion Through differential adherence and shaking at a constant temperature,more astrocytes of high purity and in good state can be obtained.

ABSTRACT

Objective To analyze the basic clinical characteristics,auxiliary diagnostic indexes,bacterial infection spectrum and drug resistance of child urinary tract infection to provide the basis for the prevention,diagnosis and treatment of child UTI.Methods The clinical data in the inpatients with UTI or complicating UTI in our hospital from January 2012 to March 2014 were collected. The repeated strains were excluded.The differences in the pathogens between the patients with complicated UTI and the patients with non-complicated UTI were comprehensively analyzed.Results The onset peak of child UTI for the first time was 0 -0.5 years old.Escherichia coli ,Klebsiella and Pseudomonas aeruginosa are the major pathogens of child UTI,in which 110 strains were Escherichia coli,accounting for 52.9%.The enterobacteriaceae pathogens of UTI had higher sensitivity to carbapenems antibacterial drugs,with the resistance rate of less than 10%;the resistance rate of others detected antibacterial drugs was more than 20%.Con-clusion UTI in the children inpatients of this area has higher drug resistance rate,in the treatment of child UTI,the antibacterial drugs should be rationally used by combining the bacterial drug resistance situation in the local place and the disease severity in or-der to avoid the aggravation of bacterial drug resistance.

ABSTRACT

Objective To understand the status of the blood safety in Handan region to provide the basis for recruiting blood do-nors in our region and the data support for meeting clinical blood use.Methods The detection results of 5 legal indicators among unpaid blood donors in Handan region during 2002-2012 were investigated and analyzed.Results The total positive rate of indica-tors detection was 5.52%,in which alanine aminotransferase(ALT)was 4.06%,hepatitis B surface antigen(HBsAg),hepatitis C virus antibody(anti-HCV)was 0.54%,human immunodeficiency virus(HIV)antibody(anti-HIV)was 0.24% and treponema palli-dum antibody(anti-TP)was 0.12%.Conclusion This survey is conducive to the blood donor recruitment work and strengthening the screening of blood donors is helpful for safety of clinical blood use.

ABSTRACT

OBJECTIVE@#To explore the effect of tanshinone IIA (TanIIA) on calcium current induced by beta-amyloid protein 25-35 (Abeta25-35) in neurons of nucleus basalis of Meynert (nbM).@*METHODS@#Cell acute dissociated technique and the whole-cell recording model of patch-clamp technique of single-cell were used. The voltage-dependent calcium current in neurons of nbM was recorded in SD rats first. Then the effect of TanIIA on the voltage-dependent calcium current in the neurons was assayed. The change of calcium current induced by Abeta25-35 as well as the effect of TanIIA on the change of calcium current induced by Abeta25-35 in neurons of nbM were analyzed.@*RESULTS@#Extracellular fluid containing different concentrations of TanIIA was irrigated, respectively. The peak current did not change obviously. There was no difference in current density between the TanIIA group and the control group at 0 mV (P>0.05). Extracellular fluid containing 200 nmol/L Abeta25-35 was irrigated after the normal calcium current recorded under whole patch clamp, and the peak current changed obviously. There was distinct difference in the current density between the Abeta group and the control group at 0 mV (P0.05).@*CONCLUSION@#In vitro, TanIIA could inhibit the calcium current amplification induced by Abeta25-35 in neurons of nbM. TanIIA may protect neurons against the toxicity of Abeta and decrease the inward flow of Ca(2+).

ABSTRACT

Objective To investigate the expression of neuroprotective peptide [Gly14]-Humanin (HNG) in eukaryotic cells by gene engineering technique and analyze its biological activity. Methods By means of asymmetrical primer/template, double stranded cDNA of HNG with FLAG in its C-terminal was obtained, which was cloned into the plasmid pcDNA3.1(-), and the resultant recombinant vector pcDNA3.1(-)/HNG-FLAG was transfected into PC12 cells. At the same time, the recombinant vector pcDNA3.1(-)/EGFP was transfected to control the efficiency of transfection. The expression of HNG in the cells was determined by immunocytochemistry. In order to analyze the biological activity of the expressed HNG, 25μM Aβ25-35 peptide was added to the culture medium of the transfected cells for 24h, then cell morphology, MTT assay and Hoechst 33258 staining were observed. Results The eukaryotic expression vector of pcDNA3.1(-)/HNG-FLAG was identified by enzyme digestion and sequencing. HNG was highly expressed in PC12 cells. After exposure of PC12 cells to 25μM Aβ25-35 for 24h, cell viability decreased to (65.8±5.3)%, and the dystrophic changes of neuritis and nuclei condensation were obvious. When cells were pre-transfected with pcDNA3.1(-)/HNG-FLAG, Aβ25-35-induced cell death and morphological changes of cells and nuclei were suppressed. In contrast, pre-transfected with empty vector did not protect cells from Aβ25-35-induced toxicity. Conclusion The eukaryotic expression vector for FLAG-tagged HNG was successfully constructed and expressed in PC12 cells. Expressed HNG has biological activity.

ABSTRACT

Objective To investigate podocyte number, the expression of nephrin and transforming growth factor-β1(TGF-β1) in adriamycin-induced-nephropathy rat model and its significance. Methods The rat adriamycin nephrosis model was constructed to detect blood and urine biochemical indicators and observe the pathological changes of renal tissues by light microscope and electron microscope. The expression levels of nephrin and TGF-β1 as well as the podocyte number were examined at different time points by immunohistochemistry. Results The pathological changes of the renal tissues were obvious. Nephrin presented a weak signal at the end of the first week (P<0.05). TGF-β1 started to increase (P<0.05) while the podocyte number started to decrease at the end of the eighth week (P<0.05). Expression of nephrin was negatively correlated with the P<0.05) and serum creatinine (r=-0.71, P<0.05). Expression of TGF-β1 was blood urea nitrogen (r=0.62, P<0.05) and serum creatinine (r=0.59, urinary protein (r=-0.63, P<0.05), blood urea nitrogen (r=-0.72, P<0.05) and serum creatinine (r=-0.76, P<0.05); it was positively correlated with nephrin (r=0.78, P<0.01) but negatively correlated with TGF-β1 (r=-0.64, P<0.05). Conclusion The acute and chronic adriamycin nephrosis models were twice every two weeks. The genesis and development of proteinuria are closely related to the abnormal expression of nephrin. Focal segmental glomerulosclerosis occurs when the podocyte number decreases and TGF-β1 accelerates it.

ABSTRACT

Objective To study the adriamycin nephrosis model in different development pathological stages. Methods The rat adriamycin nephrosis model was established by injecting adriamycin into tail-vein twice every two weeks to detect blood and urina biochemical indicators and to observe the pathological changes of the renal tissues. Results The model showed serious edema,proteinuria,hypoproteinemia,and hyperlipidemia. Podocytes were swollen and mesangial cells developed mild hyperplasia at the end of the fourth week. The nephric tubule atrophied at the end of the eighth week accompanied with adhesion between glomeruli and Bowman's capsule. Glomeruli sclerosis of mild or medium degree was observed at the end of the twelfth week with obvious lymphocyte infiltration in the renal interstitium as well as the formation of collagen fibers. Conclusion The adriamycin nephrosis model was successfully developed by injecting adriamycin 4 mg/kg into tail-vein twice every two weeks. The acute model is similar to human minimal change disease,and the chronic model is similar to human focal segmental glomerulosclerosis.
